The use case here is we have list of people whom we want to trigger an email but 'from address' and 'reply to email' address should be of their manager only which would be picked from contact list.

Unfortunately, that would not be possible unless you are using SMTP relay. If you are using SMTP and configuring it on your end, then it might be possible. But for that as well you will have to configure the from address and reply to address on your end for each person by running a query.
